β-lactam antibiotics are commonly used medication to treat bacterial infections, however certain species including Escherichia coli and Klebsiella pneumoniae, have acquired resistance through expressing β-lactamase. β-lactamase hydrolyses β-lactam rendering ineffective against populations expressing the enzyme. There are a number of genes expressing β-lactamase, many of which are located in plasmids, which cause specific resistance to certain types of antibiotics. Chromosomally located AmpC provides a low and natural resistance to β-lactam, however is generally considered to not cause the level of resistance seen in plasmid ampC β-lactamase genes. Below details genes with notable interest when expressing ampC β-lactamase:
- DHA – this gene causes resistance to oxyimino-cephalosporins and cephamycins, thought to originate from M. morganii
- CMY-2 – responsible for causing broad spectrum resistance, including ceftriaxone and ceftiofur. It has been known for organisms expressing this gene to be located in meat products and the environment, commonly found species like E. coli and S. enterica.
- ACC – this gene can be found inK. pneumoniae, Escherichia coli, and other Enterobacteriaceae.
Product Details:
The range of assays described below are in vitro PCR reaction assays for the quantitative determination of DHA, CMY-2 and ACC genes within bacterial cells. Gene expression can be measured using samples such as whole blood based on Taqman detection method for DHA, CMY-2 and ACC genes with a highly sensitive one step qPCR kit.
Quality: All AMD kits are manufactured under high-quality standardization methods with unique precision and sensitive technology when compared to the most of the famous and approved diagnostic commercial Plasmid ampC β-lactamase qPCR assays.
Specificity: AMD Plasmid ampC β-lactamase kit shows up to 100% specificity for DHA, CMY-2 and ACC genes under our validation methods and devices.
