Neisseria meningitidis, Haemophilus influenzae, and Streptococcus pneumoniae are major bacterial pathogens responsible for invasive respiratory and central nervous system infections, particularly meningitis and bacteremia.

Neisseria meningitidis is a Gram-negative diplococcus transmitted via respiratory droplets, capable of causing meningococcemia and meningitis, with virulence factors such as a polysaccharide capsule, pili, and endotoxin contributing to rapid systemic illness and high mortality if untreated.

Haemophilus influenzae, a small Gram-negative coccobacillus, can cause meningitis, epiglottitis, pneumonia, and otitis media, with encapsulated type b (Hib) strains historically being the most virulent; widespread vaccination has dramatically reduced invasive Hib disease.

Streptococcus pneumoniae, a Gram-positive, encapsulated diplococcus, is a leading cause of pneumonia, meningitis, otitis media, and sepsis, with over 90 serotypes defined by capsular polysaccharides; its virulence is mediated by factors such as the capsule, pneumolysin, and surface adhesins. All three pathogens are transmitted via respiratory droplets, and prompt recognition, antibiotic treatment, and, when available, vaccination are critical to prevent severe outcomes.
