Overview.

Tetracycline is a broad-spectrum antibiotic showing effectiveness against gram positive and gram negative bacteria, mycoplasmas, and protozoan parasites. The antibiotic prevents the production of protein synthesis through its ability to stop the attachment of aminoacyl-tRNA to the ribosomal acceptor site. However, due to its high effectiveness and inexpensive production, Tetracycline resistance is steadily on the rise. Genes like tetA and tetB cause the removal of tetracycline from the organism, preventing any protein synthesis inhibition from taking place.

Vancomycin has been the preferred antibiotic when treating infections caused by gram-positive infections and resistant to methicillin. However, resistance towards vancomycin is on the rise, but developed in the laboratories rather than due to intense prescribing of the antibiotic. Organisms like  E. faecalis and E. faecium express vancomycin resistant genes e.g. vanB.

The range of assays described below are in vitro PCR reaction assays for the quantitative determination of tetA, tetB and vanB genes within antibiotic resistant bacterial cells. Gene expression can be measured using samples such as whole blood based on Taqman detection method for tetA, tetB and vanB with a highly sensitive one step qPCR kit.
